499 miR-204-5p mimic application induces BCL2 and SIRT1 elevated expression levels in lungs and BCL2 elevated levels in kidneys of melanoma B16-bearing mice

MicroRNAs are small non-coding RNA molecules involved in the epigenetic regulation of carcinogenesis. miR-204-5p was shown to be among most downregulated miRs melanoma versus benign melanocytic lesions. SIRT1 as a gene target regulates proliferation, apoptosis, migration cancer cells whereas BCL2 is well known by it’s antiapoptotic functions. Therefore B16-bearing mice were treated synthetic mimic administered intraperitoneally once three days within two weeks. PBS used Control and scrambled antisense oligonucleotides Negative Control. overexpression resulted increase expression levels lungs (p=0.017) (p=0.043), an compared (p=0.043). Besides, increased kidneys mimic-treated animals group animals. application did not altered B16 tumors weight volume. Animals from all groups studied show any signs toxicity. Antisense oligonucleotides-based therapy triggers alterations distant organs but tumor itself that could considered for anticancer effects adjuvant treatment approaches.
